Fibroblast Biology for Antibody Discovery

Mestag Therapeutics is focused on developing innovative therapies that leverage fibroblast immunology to enhance immune responses against cancer and inflammatory diseases. Their lead product, MST-0300, is a bispecific antibody designed to induce the formation of Tertiary Lymphoid Structures (TLSs) within solid tumors. TLSs are aggregates of immune cells that play a crucial role in driving anti-tumor immune responses by recruiting and activating T and B cells. The presence of TLSs has been associated with improved patient outcomes and better responses to therapy across various solid tumor types. MST-0300 aims to conditionally activate the lymphotoxin beta receptor (LTBR) in the tumor microenvironment, promoting TLS formation and enhancing immune cell recruitment and education, thereby improving therapeutic efficacy for cancer patients.

In addition to their cancer-focused initiatives, Mestag is also pioneering the development of M402, a first-in-class stromal checkpoint agonist antibody aimed at addressing chronic inflammatory diseases. This therapy targets myeloid-driven immune responses, which are often overactive in conditions such as rheumatoid arthritis and lupus. By enhancing checkpoint receptor function, M402 seeks to dampen excessive immune activity and promote durable remission in patients suffering from autoimmune disorders. Mestag's approach is underpinned by a deep understanding of fibroblast biology and its role in immune regulation, utilizing advanced modeling techniques and multi-cellular co-culture assays to identify novel drug targets and optimize therapeutic outcomes.
