Fusogenic extracellular vesicle delivery technology

Vesigen Therapeutics has developed a novel non-viral platform that addresses significant delivery challenges associated with gene editors, proteins, and RNA. Central to this platform are ARRDC1-Mediated Microvesicles (ARMMs), which are engineered from human extracellular vesicles. These ARMMs facilitate efficient and non-immunogenic delivery of therapeutic payloads to specific tissues and cell types that have traditionally been difficult to target. By leveraging the natural biology of these vesicles, Vesigen aims to overcome limitations posed by conventional delivery methods, such as viral vectors and lipid nanoparticles, which often face issues related to biodistribution, immunogenicity, and manufacturing complexities.

The engineering process involves expressing the ARRDC1 protein along with the therapeutic payload in producer cells, which then bud off ARMMs containing the desired cargo. This streamlined, one-step synthesis allows for easy collection and purification of the engineered vesicles. Preclinical studies have demonstrated the ability of ARMMs to deliver a variety of payloads across multiple tissues, including the liver, lungs, and nervous system, showcasing their potential for treating a wide range of diseases. Furthermore, Vesigen is enhancing the targeting capabilities of ARMMs by incorporating specific surface moieties, enabling precise delivery to cell types such as hematopoietic stem cells and neurons, thus paving the way for innovative therapeutic strategies.
