NeoTX is pioneering a novel approach to cancer therapy through its Tumor Targeted Superantigen (TTS) technology, which aims to enhance the immune system's ability to recognize and attack tumors. This technology leverages the observation that the immune system is more adept at targeting bacterial cells than cancer cells. By coating tumors with superantigens—bacterial derivatives—TTS technology converts a weak immune response against tumors into a robust antibacterial response. This innovative method has the potential to improve the effectiveness of existing cancer treatments, including checkpoint inhibitors, chemotherapy, and CAR-T cell therapy, thereby offering new hope for patients who do not respond to conventional immunotherapy.

The lead candidate from NeoTX's TTS platform is naptumomab estafenatox (NAP), which is currently undergoing clinical trials for advanced solid tumors. NAP is designed to target tumors expressing the 5T4 antigen, commonly found in various cancers. Ongoing clinical trials, including a Phase 1b study in collaboration with AstraZeneca, are evaluating NAP's safety and efficacy when used in combination with checkpoint inhibitors like durvalumab and chemotherapy agents such as docetaxel. Previous studies have indicated that NAP is well-tolerated and shows promising preliminary efficacy, positioning it as a significant advancement in the field of cancer immunotherapy.
