By encoding up to 40 neoantigens in a single vaccine, Geneos aims to stimulate a robust immune response that targets all relevant mutations, rather than relying on pre-selected "high-value" neoantigens. The PTCVs are administered intradermally, accompanied by a cytokine adjuvant and electroporation, which enhances the uptake of the DNA plasmids and promotes the generation of tumor-infiltrating lymphocytes (TILs) that are crucial for attacking cancer cells.

The clinical efficacy of Geneos' PTCVs has been demonstrated in ongoing trials, particularly in patients with advanced hepatocellular carcinoma (HCC). The combination of the personalized vaccines with immune checkpoint inhibitors has shown promising results, including complete and partial responses in several patients. The platform's ability to rapidly manufacture vaccines—within six to eight weeks, with plans to reduce this to three to four weeks—ensures timely treatment for patients. Additionally, the favorable safety profile of these vaccines allows for their integration with existing standard care treatments, potentially enhancing overall therapeutic outcomes without increasing toxicity.
