Naxitamab-gqgk binds to the glycolipid GD2. GD2 is a disialoganglioside that is overexpressed on neuroblastoma cells and other cells of neuroectodermal origin, including the central nervous system and peripheral nerves. In vitro, naxitamab-gqgk was able to bind to cell surface GD2 and induce complement dependent cytotoxicity (CDC) and antibody dependent cell-mediated cytotoxicity (ADCC).

The GD2-SADA construct was created using the Company’s SADA platform, which was licensed by the Company from Memorial Sloan Kettering Cancer Center (“MSK”) and Massachusetts Institute of Technology (“MIT”). SADA utilizes a pre-targeted payload delivery method where antibody constructs assemble in tetramers and bind to the tumor target. In prior nonclinical studies unbound constructs predictably disassembled into smaller antibody fragments and were taken up by the liver or excreted through the kidneys within a few days after administration. In a second infusion, a radioactive payload designed specifically to target the SADA molecules attached to the tumor target. We believe this approach provides the possibility of targeting tumors with precision while minimizing radiation of normal tissues. We believe the SADA platform has the potential to deliver a variety of payloads and be developed against multiple tumor targets, as well as for theragnostic purposes.

A cancer vaccine, containing epitopes of the gangliosides GD2 and GD3 conjugated with the immunostimulant keyhole limpet hemocyanin (KLH), with potential antineoplastic activity. Vaccination with GD2 lactone/GD3 lactone-KLH conjugate bivalent vaccine may elicit antibodies against tumor cells expressing either epitope, resulting in complement-mediated cytotoxicity (CMC) and antibody-dependent cell-mediated cytotoxicity (ADCC). This antibody—radiolabeled monoclonal antibody 8H9 (131I-8H9)—is being developed to target B7-H3–expressing cells in human solid tumors, including embryonal tumors, carcinomas, sarcomas, and brain tumors. In vitro/ex vivo modeling has shown omburtamab binds to an FG loop-dependent conformation on the B7-H3 molecule, a domain critical for its biologic function.
 
Omburtamab is a promising investigational agent for radioimmunotherapy of leptomeningeal metastases, Diffuse Intrinsic Pontine Glioma (DIPG), and malignant ascites.
