COVID-19

NVX-CoV2373 is a protein-based vaccine candidate engineered from the &nbsp;genetic sequence of SARS-CoV-2, the virus that causes COVID-19 disease. &nbsp;NVX-CoV2373 was created using Novavax' recombinant nanoparticle Matrix-M™ adjuvant technology to generate antigen derived from the coronavirus spike (S) protein and is adjuvanted with Novavax' &nbsp;patented saponin-based Matrix-M™ to enhance the immune response and &nbsp;stimulate high levels of neutralizing antibodies. NVX-CoV2373 contains &nbsp;purified protein antigen and can neither replicate, nor can it cause &nbsp;COVID-19.
NVX-CoV2373 was shown to be highly immunogenic in animal models measuring spike protein-specific antibodies, antibodies that block the binding of the spike protein to the receptor and wild-type virus neutralizing antibodies. High levels of spike protein-specific antibodies with ACE-2 human receptor binding domain blocking activity and SARS-CoV-2 wild-type virus neutralizing antibodies were observed after a single immunization.

Novavax has developed a protein-based monovalent XBB.1.5 COVID vaccine candidate. Novavax's protein-based vaccine candidate would be the only non-mRNA XBB.1.5 vaccine available in the U.S.

NVX-CoV2705 is an updated version of Novavax's prototype COVID-19 vaccine (NVX-CoV2373) formulated to target the JN.1 variant. It is a protein-based vaccine made by creating copies of the surface spike protein of SARS-CoV-2 that causes COVID-19. With Novavax's unique recombinant nanoparticle technology, the non-infectious spike protein serves as the antigen that primes the immune system to recognize the virus, while Novavax's Matrix-M adjuvant enhances and broadens the immune response. The vaccine is packaged as a ready-to-use liquid formulation and is stored at 2° to 8°C, enabling the use of existing vaccine supply and cold chain channels.

Matrix-M™&nbsp;is Novavax’ next-generation, patented &nbsp;saponin-based adjuvant, powered by a new formulation that provides a potent adjuvant effect and is well-tolerated. Saponins are steroid or triterpenoid glycosides, which occur in many plant species. &nbsp;In Matrix-M, purified saponin fractions are mixed with synthetic cholesterol and a phospholipid to form stable particles than can be readily formulated with a variety of vaccine antigens.&nbsp; Saponin-based adjuvants act in part by stimulating the entry of antigen-presenting cells into the injection site and enhancing antigen presentation in the local lymph nodes.&nbsp;Thus, Matrix-M™ induces both a cell-mediated and an antibody mediated immune response, which we expect will be important in developing future vaccines. Matrix-M™&nbsp;also increases the opportunity for an immune reaction with longer duration, which can reduce the number of vaccinations needed to gain optimal protection.
