Nasopharyngeal carcinoma

Toripalimab was the first domestic anti-PD-1 monoclonal antibody approved for marketing in China. &nbsp;More than thirty company-sponsored clinical studies covering more than &nbsp;fifteen indications have been conducted globally, including in China and the United States. On December 17, 2018, Toripalimab obtained a conditional approval from the National Medical Products Administration (NMPA) for the second-line treatment of unresectable or metastatic &nbsp;melanoma. Toripalimab was included in the 2019 and 2020 Guidelines of Chinese Society of Clinical Oncology (CSCO) for the Diagnosis and Treatment of Melanoma. The supplemental NDA of Toripalimab for the second-line &nbsp;treatment of metastatic urothelial carcinoma was accepted by the NMPA in &nbsp;May 2020 and received priority review designation from the NMPA in July 2020. In September 2020, Toripalimab was granted Breakthrough Therapy Designation by the US Food and Drug Administration (FDA) for the treatment of recurrent or metastatic nasopharyngeal carcinoma. In December 2020, Toripalimab was successfully included in the updated National Reimbursement Drug List. In February 2021, &nbsp;the supplemental NDA for Toripalimab in combination with chemotherapy &nbsp;for the first-line treatment of patients with advanced, recurrent or &nbsp;metastatic nasopharyngeal carcinoma was accepted by the NMPA. In the &nbsp;same month, the NMPA granted a conditional approval to toripalimab for &nbsp;the treatment of patients with recurrent or metastatic nasopharyngeal &nbsp;carcinoma (NPC) after failure of at least two lines of prior systemic &nbsp;therapy. Currently, Toripalimab has been granted 1 Breakthrough, 1 Fast &nbsp;Track, and 3 Orphan Drug Designations by the FDA for the treatment of &nbsp;mucosal melanoma, nasopharyngeal carcinoma, and soft tissue sarcoma.

YUSIMRY (CHS-1420) is a US FDA approved biosimilar of HUMIRA (also known as adalimumab). Adalimumab binds specifically to TNF-alpha and blocks its interaction with the p55 and p75 cell surface TNF receptors. Adalimumab also lyses surface TNF expressing cells in vitro in the presence of complement. Adalimumab does not bind or inactivate lymphotoxin (TNF-beta). TNF is a naturally occurring cytokine that is involved in normal inflammatory and immune responses. INO-3112 is a DNA medicine candidate targeting HPV 16/18 combined with a DNA plasmid for IL-12 as an immune activator. INOVIO is investigating the potential benefit of the antigen-specific T cell generation and tumor infiltration abilities of INO-3112 in HPV-related cancers, especially when used in novel combinations. Results from a Phase 1/2a trial of INO-3112 alone in 22 HPV-positive head and neck squamous cell carcinoma (HNSCC) patients, published in Clinical Cancer Research in 2019, included the observation of T cell responses and&nbsp;infiltration of CD8+&nbsp;T cells into the head and neck&nbsp;tumors. In early 2023, updated results were published in Clinical Cancer Research from a Phase 1b/2a trial of INO-3112 in combination with AstraZeneca's PD-L1 checkpoint inhibitor, durvalumab, showing an ORR of 27.6% (4 CR, 4 PR) in 29 evaluable patients and increased peripheral HPV-specific T cells and tumoral CD8+ T cells.

Toripalimab (CHS-305) is a US FDA approved biosimilar of bevacizumab. Bevacizumab binds VEGF and prevents the interaction of VEGF to its receptors (Flt-1 and KDR) on the surface of endothelial cells. The interaction of VEGF with its receptors leads to endothelial cell proliferation and new blood vessel formation in in vitro models of angiogenesis.
Administration of bevacizumab to xenotransplant models of colon cancer in nude (athymic) mice caused reduction of microvascular growth and inhibition of metastatic disease progression.
