Synovialsarcoma

Afami-cel is an engineered T-cell receptor (TCR) T-cell therapy, targeted to the MAGE A4 cancer target, and designed as a single-dose treatment for advanced synovial sarcoma. The last FDA approved therapy for treatment in this setting was for Votrient in 2012. The BLA submission for afami-cel was supported by clinical data from the SPEARHEAD-1 pivotal trial, which has met its primary endpoint for efficacy. ~39% of patients who received afami-cel had clinical responses with a median duration of response of ~12 months (<a href="https://api.newsfilecorp.com/redirect/oLq7xuwzbJ">CTOS 2022</a>). Median overall survival (mOS) was ~17 months in SPEARHEAD-1 compared to historical mOS of &lt;12 months for people with synovial sarcoma who received two or more prior lines of therapy.1 Seventy percent of people with advanced synovial sarcoma who respond to afami-cel are alive two years post-treatment.
 
<h3 class="font_3">Cell Manufacturing:</h3>
We are enhancing the body’s natural immune defenses to specifically target solid tumor cancers by optimizing the binding of T-cells and their receptors such that they can recognize cancer cells and clear them from the body.
Once we have affinity-enhanced a TCR, we can deliver it to patients through a robust and commercialized manufacturing process. To manufacture our TCR therapeutic candidates, we collect blood cells from a cancer patient using a process called leukapheresis. CD4 (helper T-cells) and CD8 (cytotoxic T-cells) cells are isolated from the white blood cells. We then deliver the genes encoding the affinity-enhanced TCRs to the T-cells.
These affinity enhanced T-cells are then expanded through a process using DynaBeads® CD3/CD28 beads which provide healthier cells and lead to robust expansion compared to other validated expansion methods. The cells are then concentrated and frozen until the patient is ready to receive them.

Lete-cel is being investigated for the treatment of synovial sarcoma and myxoid/round cell liposarcoma (MRCLS) in the pivotal IGNYTE-ESO (NCT03967223) trial in patients who received prior anthracycline treatment.

Our ADP-A2M4CD8 SPEAR T-cell therapy is our first "next-generation" therapy and is directed to MAGE-A4, a member of the MAGE family of cancer-testis antigens expressed in a number of solid tumor types. What differentiates this therapy from our ADP-A2M4 SPEAR T-cells is that these cells also express the CD8α co-receptor alongside the engineered TCR that targets MAGE-A4. Preclinical data indicate that co-expression of CD8α may broaden the immune response against solid tumors and increase antitumor activity by leveraging CD4+ cells into CD8+ killer or cytotoxic T‑cells while retaining their CD4+ helper function.

Our ADP-A2M4 (MAGE-A4) SPEAR T-cell therapy is directed to a member of the MAGE family of cancer testis antigen expressed in a number of solid tumor cell types. The MAGE- A4 antigen is among the most commonly expressed cancer-testis antigens. We are conducting various trials with the ADP-A2M4 SPEAR T-cell in various solid tumors.

TCRR are developing TC-210 to target mesothelin-positive solid tumors. While its expression in normal tissues is low, mesothelin is highly expressed in a wide range of solid tumors and it plays an active role in both malignant transformation and tumor aggressiveness by promoting cancer cell proliferation, invasion, metastasis. Mesothelin overexpression has also been correlated with poorer prognosis in certain cancer types.

We are developing TC-510 to target mesothelin-positive solid tumors. The PD-1:CD28 switch receptor built into this TRuC-T cell is designed to convert the PD-L1/2 inhibitory function into a potent costimulatory signal. Costimulation occurs only in a PD-L1/2 rich tumor microenvironment upon TRuC and PD-1 ligation, resulting in a more targeted signal enhancement.
