Therapies for the treatment of Idiopathic Pulmonary Fibrosis

Avalyn Pharma is focused on developing innovative inhaled therapies for rare respiratory diseases, particularly interstitial lung diseases (ILDs) such as pulmonary fibrosis. Their lead candidate, AP01, is an inhaled formulation of pirfenidone, which is designed to target the lungs directly, thereby maximizing therapeutic effects while minimizing systemic side effects associated with oral administration. This reformulation utilizes an approved handheld eFlow® vibrating membrane nebulizer to deliver small doses of the drug as a soft mist, allowing for effective treatment of the lung tissue without the adverse gastrointestinal effects commonly seen with oral pirfenidone.

In addition to AP01, Avalyn is also developing AP02, an inhaled formulation of nintedanib, which targets multiple tyrosine kinases involved in lung fibrosis. This inhaled version aims to reduce the systemic side effects associated with oral nintedanib, such as diarrhea and liver injury, by delivering the drug directly to the lungs at significantly lower doses. Furthermore, Avalyn is exploring a fixed-dose combination product, AP03, that combines both pirfenidone and nintedanib for inhaled delivery, potentially enhancing efficacy while reducing side effects. This innovative approach to drug delivery is expected to improve patient compliance and overall treatment outcomes for those suffering from these debilitating conditions.
